@Ryan R if you put the nmos inside a pwell (for a triple well isolation - that is, pwell inside deep nwell, surrounded by nwell, inside the p substrate), then you have to draw the pwell GDS layer. Otherwise, the p substrate (which has a different P dopant concentration from the p well) is all you need - no extra GDS layer
