Is there are mechanical / DRC / yield / ... issue if using a metal layer for a long track in a non-preferred direction ?

Ok, yeah. As far as I am aware that's just to help the routing. There's no physical requirement for that. Analogue designs for example will quite often break those conventions

That changes when you get to much smaller nodes where it's much easier to lithograph straight lines than arbitrary shapes so I believe there are real rules there but not in the sky130 node
